In the original HoMM5 it isn't possible, but in one of the expansions (I think it was Tribes of the East) a building was added that allowed a hero to unlearn skills (for a cost of gold). You'd then be able to immediately re-assign any removed skills, so it was basically the ability to respec as you saw fit.
